• We proposed an algorithm to predict new drugs for diseases based on tissue specificities and the triangularly balanced structure on incomplete data.

• Taking breast cancer and HCC as case studies, 96.9% and 90.3% in the top-5% predicted results match with known associations in CTD respectively.

• We predicted: Pralatrexate may be a potential treatment for breast cancer; Vindesine and Cabazitaxel may be treat HCC; Pralatrexate may be cause HCC.
